Oligosaccharides found in beer were separated by gradient elution in around 45 minutes by reversed phase HPLC on a Prevail Carbohydrate ES, 5µm, 250 x 4.6mm column (Part No. 35101) using acetonitrile/water and detected by evaporative light scattering using an Alltech 2000 ELSD (Evaporative Light Scattering Detector).
Frucotose, glucose, maltose, maltotriose, maltopentaose and maltohexaose were separated in New Castle Ale and maltotriose, maltohexaose and maltoheptaose were separated in Goose Island Beer.
In ELSD, the mobile phase is first evaporated. Solid particles remaining from the sample are then carried in the form of a mist into a cell where they are detected by a laser.
