The ELSD and UV detection of five fatty acid methyl esters separated by reversed phase HPLC was compared.

The ELSD and UV detection of five fatty acid methyl esters (FAME) separated by reversed phase HPLC was compared. The FAME compounds were separated isocratically in around 10 minutes by reversed phase HPLC on a Silver Impregnated Ion-Exchange, 250 x 4.6mm column (Part No. not given) using acetonitrile/methanol and detected by evaporative light scattering (ELSD) using an Alltech Model 500 (Evaporative Light Scattering Detector) and by UV at 206 nm.
All five of the FAME compounds (18:0, 18:1 n-9, 18:2, n-6, 18:3 n-3 and 20:4 n-6) were resolved by ELSD, but only the last 3 were resolved by UV.
In ELSD, the mobile phase is first evaporated. Solid particles remaining from the sample are then carried in the form of a mist into a cell where they are detected by a laser.
