The chiral separation of d and l forms of warfarin was performed by reversed phase HPLC using isocratic elution in around 6 minutes on an Astec Cyclobond 1 2000, 5µm, 250 x 4.6mm column (Part No. 410101) using acetic acid/TEA and acetonitrile/methanol. Detection was achieved both UV at 254 nm and by evaporative light scattering using an Alltech 500 ELSD (Evaporative Light Scattering Detector).
In ELSD, the mobile phase is first evaporated. Solid particles remaining from the sample are then carried in the form of a mist into a cell where they are detected by a laser.
