MMDA/MDA (Ecstasy) Determination by GC and HPLC

MDMA (3,4-methylenedioxymethamphetamine), an illegal drug also known as Ecstasy, and several of its analogs were separated by both GC/FID and HPLC/UV.

MDMA (3,4-methylenedioxymethamphetamine), an illegal drug also known as Ecstasy, and several of its analogs were separated by both GC/FID and HPLC/UV.

The GC separation was accomplished on an Alltech Heliflex® ATTM-5ms, 15m x 0.25mm x 0.25µm capillary column (Part No. 15801) isothermally at 160°C using helium carrier gas. Detection was by FID at a temperature of 300°C. The separation of 3,4-MDMA, 2,3-MDMA, 2,3-MDA and 3,4-MDA took about 4 minutes.

The HPLC separation of these same four compounds in less than three minutes was accomplished on a Platinum® Silica, 3µm, 100 x 4.6mm reversed phase column (Part No. 32524) using a mobile phase of (75:25) acetonitrile/1% ammonium hydroxide in methanol and a flow rate of 2.0mL/min. Detection was by UV at 280nm.
